Bleomycin Induced Chromosome Rearrangement Detected by Fluorescence in situ Hybridization Using Triple Combination of Composite Whole Chromosome-specific Probes in Human Lymphocyte

Abstract
Chromosome rearrangements induced by bleomycin were identified by fluorescence in situ hybridization (FISH) with triple combination of composite whole chromosome-specific probe for chromosome 1, 2 and 4. Triple combination of chromosome-specific probe covers larger proportion of DNA content in the whole genome, and higher validity in scoring chromosome rearragements can be achieved than the single chromosome-specific probe. Bleomycin treatment resulted in a dose dependent increase of all type of chromosome rearrangements scored by Protocol for Aberration Identification and Nomencluture Terminology (PAINT) System. Translocation was more frequently observed than dicentric chromosomes. And Bleomycin treatment has resulted in complex exchanges and cells with multiple aberrations.
